What is the GnomePen Classic

The GnomePen Classic is a roller PAP pen applicator. It contains 7ml of our hydrophobic blocker which, depending on usage, suffices for approximately 300 slides.   Our unique PAP Pen formulation average drying time is very fast – only 1 minute on the bench and under 30 seconds in a chemical fume hood – faster than any competing product. 

Our PAP Pen formulation adheres both to plain glass slides, Silan-subbed slides, plastic and even to oil-covered glass slides (as usually supplied directly from the factory). However, it will not adhere to wet slides. Therefore, we recommend applying the GnomePen Classic hydrophobic pen as the very first step in the IHC or IFA protocol. For paraffin embedded samples – the Gnomepen should be applied after removing the paraffin – simply wipe a ring around the slice using a Q-tip and apply the GnomePen in the semi-dry area. 

Hydrophobic blocker formulation

Our PAP Pen hydrophobic barrier liquid forms a thin layer on the glass slide which can be removed using Xylene or even manually with a tip or a q-tip. However this is rarely required as the hydrophibic barrier line is not tall. Hence, the coverslip can be applied directly over the hydrophobic barrier line. 

Unlike regular PAP Pens our formulation is fixative and detergent resistant for long durations. Paraformaldehyde, formadehyde (formalin) fixatives and detergents like Triton X100, X114 and Tween 20 do not affect the hydrophobic blocker. Our formulations will not interfere with most antibodies, flourophores or reagents. 

Supplied in six colors the GnomePen Classic enables our customers to color code their work: color coding different slices, antibodies and most importantly – to distinguish  experimental slices from the control slices. OurTip: use a red GnomePen for your negative control and you will not turn it into a positive one! 

Last but not least – for special finicky antibodies – ask for our Clear PAP Pen formulation. Upon request (at the same price) a clear hydrophobic blocking formulation can be supplied.
