Where to start? First of all – Consult your PI. He knows it, he has done it, and if not – he will know who to reffer you to. If you have a laboratory manager – that’s the person to consult with too! Ask around and don’t be shy! Ask your colleagues within and outside your laboratory. Be sure to especially ask the postdocs: they have done a LOT of scientific work so far.

Choose your protocol Look on PubMed for highly cited articles in your field of expertiese – their protocols have been tested by more than one laboratory and one person. This means they are more reliable and can produce better and more consistent results.

Choose your samples If you are just starting – opt for absolutely positive samples that will surely contain your proteins or peptides. Start with those and see how it goes. Do not start with an unknown sample – you will not know if it’s the sample that is negative or your protocol or antibodies that fail you!

Experiment THE best teacher is someone who has done it before. Find a laboratory next to your location that actually does it. You need help from someone that actually is cutting tissues either on a microtome or cryotome (a frozen micrometer) and does a routine immunohistochemistry or immunofloursecense. Ask for their help – and most likely they will agree. Join them, see what they do and write notes. Better yet – ask for their premission to run an expreiment in their laboratory or join their experiment and run the protocol yourself!
